Amsterdam and Brussels are of the same size with a small advantage to Amsterdam.

Amsterdam is a famous city and will more easily attract tourists to the city as Brussels is more like a business centre with the European union and all that. That is why you will find more easily pure tourist hotels in Amsterdan Netherlands. Of Course hotels in Brussels Belgium are possible more expensive and more luxurious because of the easily spent money of corporations but charm is not going hand in hand with this development.

If i would personally had to choose between a weekend in Amsterdam or a Weekend in Brussels i would easily choose a hotel Amsterdam because of the richer historical sites and museums. Amsterdam has still preserved the old layout of the city and has not had the hugh change Brussels underwent the moment it became the capital of Belgium.

Many streets and original layout was destroyed to make place for modern buildings, schools, palaces and academies. In Amsterdam, all of that was integrated into the city without destroying the older historical buildings.

Brussels, The European Capital, ...

Depending on how much you want to spend, one can easily find a hotel which is perfect for hir of her budget.
Public transportations to the airport:

Trains: You can fetch a train in Brussels South, Central or North. Every 10 minutes you will have a train leaving to the airport. It takes about 20 minutes to arrive. The trainstation in Brussels National Airport is situated beneath the arrival and departure halls.

Car: Risky but managable on certain hours of the day. From the inner city it normally should not take more than 30 minutes to reach the airport but rush hour can be a bit troubeling.

Bus: Rather not.

Metro: you have to take the train but metro stops close all three trainstations.

Distance to other cities who are definitely worth visiting: Bruges (1h in train), Ghent (30min in train), Anwerp (40 min in train ) and perhaps the seaside Ostend (1.30 form brussels in train ).
